组织芯片技术原位检测石蜡包埋肝细胞癌组织的研究

【摘要】 目的 探讨组织芯片技术高通量原位免疫组织化学检测石蜡包埋肝细胞癌组织的价
值。方法 1991 年1 月- 2002 年6 月西南肝胆医院实施根治性肝切除,病理证实为肝细胞癌234 例
石蜡组织块,健康成人肝组织18 例(西南医院肝胆外科研究所肝移植供肝组织) 作为对照,重新包埋
石蜡组织,制备成为2 个微阵列,分别为13 ×10 (129 点) 和14 ×9 (123 点) ,微阵列点直径1. 0 mm ,点
间距为0. 5 mm。Envision 法检测Vimentin 和E2cadherin 的表达。结果 组织芯片中组织样本的组
织学结构完整,可满足病理学实验要求。结论 组织芯片技术提供了一种快速、有效的组织学方法,
可广泛应用大样本肝细胞癌的免疫组织化学检测。

【Abstract】 Objective To explore the application and values of paraffin2embedded hepatocellular
carcinoma ( HCC) tissue microarray technique for high2throughput in situ immunohistochemical stud2
ies. Methods Paraffin2embedded tissue f rom 234 HCC and 18 healthy liver tissue samples f rom J anu2
ary 1991 to J une 2002 at Southwest Hospital were used for the microarray. Two TMA blocks were
const ructed. 129 specimens were arrayed on one block and 123 on the other. Core tissue biop sies of
1. 0 mm in diameter were taken f rom carefully selected morphologically representative regions of indi2
vidual paraffin2embedded samples ( donor blocks) and precisely arrayed into a new recipient paraffin
block using a custom built inst rument . Immunohistochemist ry Vimentin and E2cadhenrin expression in
HCC was determined using immunohistochemist ry technique. Results The histological st ructure of
the specimens was complete in tissue microarray. It can meet the requirement s of pathological experi2
ment s. Conclusions Tissue microarray provides a rapid and powerful histological method that may be
used for high2throughput in situ studies and play an important role in tumor biology.
